The Mouse Metabolic Phenotyping Center
at the Yale University School of Medicine

 

In Vivo Metabolism Core

In Vivo Metabolism Core

The In Vivo Metabolism Core at the Yale Mouse Metabolic Phenotyping Center is designed to conduct in vivo experiments and analytical assays to characterize the metabolic phenotype of transgenic/knockout mouse models potentially useful for understanding diabetes, its complications, obesity, and related metabolic disorders.

The Core is directed by Dr. Gerald Shulman, Professor of Medicine, and Cellular & Molecular Physiology, and Co-Directed by Dr. Varman T. Samuel, Assistant Professor in the Department of Internal Medicine. Laboratories are located in the Anlyan Center for Medical Research & Education and the Sterling Hall of Medicine of the Yale University School of Medicine.

 

The principle functions of the Core are as follows:

  1. Hyperinsulinemic-euglycemic clamp experiments to assess in vivo insulin action, insulin signaling, and glucose metabolism in awake mice.
  2. Hyperglycemic clamp experiments to assess in vivo pancreatic b-cell function (i.e., glucose-induced insulin secretion) and the effect of hyperglycemia on glucose metabolism (i.e., glucose toxicity).
  3. Mouse surgery to establish chronic cannulation of jugular vein for infusion during experiments.
  4. Implementation of treadmill to assess the effect of exercise on glucose metabolism.
  5. Performance of biochemical and radioactive analysis to estimate in vivo glucose metabolism and insulin signaling.
  6. Measurement of body fat composition in conjunction with NMR Core.
